I have no input on places to stop. I am sure you and your wife will have a great time. The farthest we ran was 411 mile from Pa to Laconia and loved it.

As a bicyclist, thread titles like this always leave me wondering. How many days do you want to take getting down there? St Augustine is about half way, and I think it's a really cool town. If you would be interested in snorkeling or seeing and doing cool stuff in the Keys let me know.

Just got back 2 weeks ago from a Daytona Beach, Key West ride. Our daughter is ill and my wife couldn't go but she insisted I go and thaw out from the long winter.
If you decide to ride down the East coast on 95 take rte 520 west at Cocoa Fl. About 8-10 miles look for a small sign for St. Cloud and turn left. This is Nova Rd. Go about another 10 miles and take the first left you can. I think its rte 419 but I know its the first left possible.
Run this to the end and take a right on rte192. Continue about 15/20 miles and you'll come to an intersection with a flashing yellow light hanging over the road. This is Holopaw and you want to turn left on rte 441. This will take you down the east side of Lake Ocochobee. Stay on 441 to Southport where you will take rte 27 on South which will eventually connect to rte1 which you will take on to Key West.
This inland route will allow you a quiet peaceful ride while eliminating the terror filled ride through West Palm Beach, Ft. Lauderdale, Boca, and Miami at rush hour. By the way, rush hour seems to be every hour.
Take it from me and just take your time and relax. Because my wife wasn't with me I felt guilty leaving her at home and I rushed through everything and did nothing more than get in a good long ride and a terminal case of monkey butt.
Coming back off the Keys I would suggest rte 41 to Naples. Get back inland heading North from Naples. Check your map but I think its rte 17 . I know you don't want rte 75.
By the way, while in Key West I highly recommend the Conch Republic Seafood
Co. for great food. Best steak I've had in a long time. I think the address is Elizabeth and Green streets.
Anyway ride safe and enjoy

I did it from south MS to Key West a couple of years ago. We took I10 to St Augustine and then south down Highway 1/A1A to Miami. Nice ride south, lots of places to stop.

What route are you taking? Most go down west side of Florida down rt 19. Gets into more traffic about the time you reach Hudson FL, but until then you see Tallahassee,Wachula Spgs. , Cedar Key,FL, Crystal River, tons of stuff. You can keep going into Tarpon spgs great Greek heritage, Dunedin, Clearwater beach, st. Pete, fort Disoto, Bradenton, Naples, alligator alley, and this is if you just stick to the west coast. Going back you could take longer and go up the east coast a1a, ft. Lauderdale, palm beach, Daytona, ormond beach St. Augustine, fernandino beach, then cutback over. Given the time a million sites.
